With FC Twente not in action until Saturday, the win for Fred Rutten's men takes them to the top of the table for 24 hours at least.
Danko Lazovic set PSV on their way to three points with the opening goal on 29 minutes but Sparta came back strongly.
Rydell Poepon equalised for the home team after 29 minutes and then grabbed his second of the evening to make it 2-1 midway through the second half.
PSV drew level at 2-2 courtesy of a Jonathan Reis effort on 69 minutes and Balazs Dzsudzsak then fired home the winner two minutes later.
The result leaves Sparta in ninth place, while PSV will stay top if Twente fail to beat Willem II.
